大鼠急性肺血栓栓塞后VEGF、HIF-1的表达变化及相关性

Changes of VEGF and HIF-1 expressions in rats with acute pulmonary embolism and their correction

  • 摘要: 目的 研究大鼠急性肺栓塞模型肺组织中血管内皮生长因子(VEGF)、缺氧诱导因子1(HIF-1)的表达变化,以了解PTE与VEGF、HIF-1α相关性。方法 建立大鼠急性肺栓塞模型,分别在急性肺栓塞后1、6、12、24h开胸取肺组织。常规提取肺组织的总蛋白,以正常组为对照,免疫组织化学法测定VEGF、HIF-1α表达水平的变化。结果 在大鼠急性肺栓塞后,HIF-1和VEGF蛋白水平均逐渐升高,在6h升高最为明显。结论 HIF-lα在大鼠PTE肺组织中呈高表达,并调节其下游基因VEGF的表达增强,可对肺组织产生保护作用。

     

    Abstract: Objective To study the changes of VEGF and HIF-1α expression in a rat model of acute pulmonary embolism(PE) and their correction. Methods Lung tissue samples were collected l,6,12 and 24h after a rat model of acute PE was established.Total protein was extracted from the lung tissue samples.Normal rats were used as controls.Immunohistochemical method was used to examine changes of HIF-lα and VEGF expression in lung tissue samples. Results The VEGF αnd HIF-1α expression levels were gradually increased after acute PE and reached their peak at 6h after acute PE. Conclusion The expression of HIF-l is up-regulated in rat lung tissue after acute PE and increases the expression of VEGF,thus protecting lung tissue from damage.
