乳腺癌干细胞模型的筛选

Screening of breast cancer stem cells model

  • 摘要: 目的:研究不同细胞系中乳腺癌干细胞的比例,为临床根治肿瘤和研发靶向性治疗药物提供理想的乳腺癌干细胞模型。方法:采用流式细胞仪对乳腺癌细胞亚系MCF7、SKB r3和人胚肾293细胞中CD44 CD24的表达情况进行检测;同时对化疗前后各细胞CD44 CD24的表达进行比较。结果:CD44+CD24-细胞在MCF7、SKBR3及人胚肾293细胞中的比例分别为0.20%±0.10%;1.37%±0.20%;36.27%±3.44%;CD44+CD24-细胞在293细胞中所占比例最多。化疗后CD44+CD24-在293细胞中比例升高明显,而在MCF7、SKBR3细胞中CD44+CD24-细胞的比例未发生明显改变。结论:MCF7、SKB r3、293细胞均存在乳腺癌干细胞,而人胚肾293细胞是临床开展乳腺癌治疗的理想乳腺癌干细胞模型。

     

    Abstract: objective: To study proporation of breast cancer stem cells in MCF7、 SKBR3 and 293 cells in order to provide a good cell model for clinical treating breast cancer and developing target drugs.Methods: To measure the expression of CD44 and CD24 in MCF7、 SKBR3 and 293 cells by flow cytometry.And to study the expression change of CD44 and CD24 after chemotherapy.Results: The expression proporation of breast cancer stem cells in MCF7、 SKBR3 and 293 cells was 0.20%±0.10%,1.37%±0.20% and 36.27%±3.44%,respectively.The expression proporation was increased from 36.27%±3.44% to 41.32%±1.06% in 293 cells after chemotherapy,and there were no obvious changes in MCF7 and SKBR3 cells.Conclusion: Breast cancer stem cells exist in MCF7、 SKBR3 and 293 cells.And 293 cells is good as breast cancer stem cells model.
