心肌肌凝蛋白轻链-1的免疫组化研究

  • 摘要: 为了研究心肌肌凝蛋白轻链-1(CMLC-1)组织特异性的特点,并对其构型在心肌发育过程中的表达规律以及不同哺乳动物CMLC-1构型的进化规律进行探讨,应用抗人心室肌CMLC-1单克隆抗体(McAB)对成人和不同胎龄胎儿的心肌和其它组织、不同种属哺乳动物的心肌进行了免疫组织化学研究。结果发现:成人心室肌,慢性骨骼肌均发生强烈反应,成人心房,胎儿心室肌发生交叉反应,而其它组织如快型骨骼肌、平滑肌以及肝脾等非肌组织不发生反应。胎儿心肌的交叉反应程度随胎龄增加而增强,哺乳动物心肌均发生交叉反应,且随动物进化等级的提高呈增强趋势。结果说明:人CMLC-1具有一定的组织特异性,CMLC-1的表达与心脏发育和动物进化程度相关。

     

    Abstract: To investigate the issue specifical characterics of human cardiac myosin light chain-1 (CMLC-1),the changes in CMLC-1 isoforms expression during the development and the evolutionary relationships of the CMLC-1 isoforms in different species of mammalian animals, we examined CMLC-1 in adult and fetal human hearts and other tisssues and mammalian hearts immuohistochemically by using of the monoclonal antibody against human ventricular light chain-1 (MabVLC-1). The results showed the adult human ventricle and slow-type skeletal muscles were strongly crossreacted with MabVLC-1, and the human adult atria and fetel ventricle were moderately crossreacted with MabVLC-1;and other human tissues including fast type skeletal muscle, smooth muscle and liver or spleen were not crossreacted. The degree of the fetal ventricle crossreacted with MabVLC-1 increaced with advance of the fetal ages. The heart tissues of all kinds of mammalains were crossreacted with MabVLC-1,and the reactivity increaced with the evolutionary degree of the mammlain species. The results suggest that (1) The CMLC-1 express specially in the ventricl and slow-type muscles;(2)The expression of the CMLC-1 in ventricle is developmentally and evolutionaryly getting move.
