诺果宁、培高利特和降乳散对乙菧酚诱发的大鼠泌乳素瘤防治作用的初步研究

The preventive effects of Norprolac, Pergolide and Jiangrusan on Diethylstilbestrol induced rat prolactinoma

  • 摘要: 目的: 研究多巴胺受体激动剂诺果宁、培高利特及中药汤剂降乳散对乙酚(DES)诱发的大鼠泌乳素(PRL)瘤的防治作用。方法: 成年Wistar大鼠切除卵巢后随机分5组,用空白硅胶管皮下植入作为对照组;其它4组均植入DES硅胶管,其中有3组分别用诺果宁、培高利特和降乳散灌胃。8周后处死动物,做垂体的免疫组化检查,用放免法测定外周血PRL水平。结果: DES可使血清PRL水平、垂体重量和PRL阳性细胞计数明显增高(P<0001)。结论: 这三种药物对DES诱发的大鼠PRL瘤均具有一定程度的防治作用。下丘脑多巴胺水平的降低可能是大鼠PRL瘤的致病因素之一。

     

    Abstract: Objective: To study the preventive effects of chinese medicine, Jiangrusan, and two kinds of dopamine receptor agonists, Norprolac and Pergolide, on Diethystilbestrol (DES) induced rat prolactinoma. Methods: forty six adult Wistar rats, ovariectomized, were divided into 5 groups at random. Each rat in E (n =10), D1n =9), D2n =10) and D3n =7) group was subscutaneously implanted with a DES implant. Rats in control group (C group, n =10) were implanted with blank implants. Meanwhile Norprolac, Pergolide or Jiangrusan was orally adimnstrated to rats in D1, D2 or D3 groups respectively. After 8 weeks, all the animals were executed. Each pituitary gland was weighed and collected for histopathological and immunocytochemical examination. Serum prolactin (PRL) levels were measured by RIA method. Results: The weights of pituitary gland, PRL immunoreactive cell countings and serum PRL levels in E, D1, D2 or D3 group were individually greater than those in C group (P <0 001 respectively). Conclusion: Norprolac, Pergolide and Jiangrusan all have potential prevenive effects on estrogen induced rat prolactinoma. The decrease of dopamine contents in hypothalamas may be involved in the pathogenesis of estrgen induced rat prolactinoma.
