Effects of RGD-containing peptide on the signalling molecule focal adhesion kinase in glomerular mesangial cells

  • 摘要: 目的:为了探讨抗粘附精-甘-天-丝(RGDS)四肽调节人肾小球系膜细胞(GMC)生物学行为的信号传递机制。方法:本实验采用体外细胞培养和逆转录-聚合酶链反应(RT-PCR)技术,观察了RGD多肽对培养GMC的生长形态、以及胞浆信号传递分子局灶粘附激酶(FAK)基因表达的影响。结果:RGD多肽能够抑制GMC与细胞外基质粘附,使GMC发生脱壁、凋亡;在此过程中,FAK的mRNA表达发生下调(Down-regulation)。结论:FAK信号分子对GMC的粘附及生存可能具有重要的调节作用。

     

    Abstract: Objective:To investigate the signalling mechanism of anti-adhesive RGD-containing peptide regulating the biological behaviors of human glomerular mesangial cells (GMCs).Methods:The methods of cell culture and reverse-transcription-polymerase chain reaction (RT-PCR) were applied to observe the changes of cellular morphology and the expression of cytoplasmic signalling molecule focal adhesion kinase (FAK) in GMCs. Results:RGD peptide could inhibit the adhesion between GMCs and extracellular matrix triggering the detachment and subsequent apoptosis of GMCs, during the process of which FAK mRNA expression was found to be down-regulated.Conclusion:Our results suggest that the signalling molecule FAK may have an important role in regulating the adhesion and survival of GMCs.
