Effect of interleukin-1 on the cytosolic Ca2+ of rat thyroid FRTL-5 cells

  • 摘要: 目的:利用激光扫描共聚焦显微镜(LSCM)观察IL-1β及TSH对鼠甲状腺FRTL-5细胞内Ca2+的影响,以进一步了解IL-1β对甲状腺细胞的作用及机制。方法:将待测FRTL-5细胞以Fluo-3负载,以LSCM扫描测定细胞内Ca2+,分别加入不同浓度的TSH或IL-1,观察细胞内Ca2+变化。结果:(1)TSH可使细胞内Ca2+的水平上升,与TSH浓度有关。(2)IL-1β可降低FRTL-5细胞内基础Ca2+和TSH刺激的细胞内Ca2+的水平。结论:TSH对甲状腺细胞磷酸肌醇/钙离子系统有刺激作用,Ca2+作为第二信使可介导TSH的效应。I-1β可以通过抑制甲状腺细胞磷酸肌醇/钙离子系统而发挥其抑制效应

     

    Abstract: Objective: In order to investigate the effect of IL-1 β on the inositol phosphate (Inos)/Ca2+ cascade in thyroid cells, the changes of cytosolic Ca2+ induced by IL-1 β and TSH were observed by laser scanning confocal microscope(LSCM) in rat thyroid FRTL-5 cells.Methods: The cultured FRTL-5 cells loaded with Ca2+ probe Fluo 3 were exposed to the different concentrations of TSH or IL 1β and then the cytosolic Ca2+ were measued by LSCM. Results: The intracelular Ca2+ levels of FRTL-5 cells were increased after adding TSH(1 to 5U/L) by dose dependent manner; while IL 1β decreased the basal levels and the TSH stimulated effect of cytosolic Ca2+ . Conclusion: TSH may elevate cytosolic Ca2+ levels, indicating that Ca2+ can mediate the action of TSH on thyroid cells; IL-1β has a direct inhibitory effect on Inos/Ca2+ cascade in thyroid cells.
