我国小型猪品系胰岛素受体底物1基因外显子1的多态性分析

Polymorphism of insulin receptor substrate 1 gene exon 1 in three Chinese native miniature pig strains

  • 摘要: 目的 探讨我国特有的三个小型猪品系—中国农大小型猪、五指山小型猪、广西巴马小型猪胰岛素受体底物1基因多态性分布,为这些小型猪在代谢性疾病和2型糖尿病研究中的应用提供基础资料。方法 提取三个品系小型猪血液基因组DNA,针对胰岛素受体底物1基因外显子1的部分序列进行PCR扩增,对产物进行鉴定和测序,分析基因多态性。结果 在所扩增的片段中共检测出2个SNPs,第2853位,T转换为C;第2870位,C颠换为G,导致第956位的氨基酸由丙氨酸(Ala)变为甘氨酸(Gly)。结论 本实验检测出的两个SNPs均发生在中国农大小型猪中,所检测的五指山小型猪和广西巴马小型猪未发现SNPs,这可能是因为小型猪品系之间存在遗传差异所致。

     

    Abstract: Objective To explore the polymorphism of insulin receptor substrate 1(IRS-1)in three strains of Chinese native miniature pigs(Zhongguonongda minipigs,Wuzhishan minipigs and Bama minipigs)in order to provide basic data for its use in metabolic disease and type 2 diabetes mellitus. Methods Genomic DNA was extracted from three strains of Chinese native miniature pigs,certain sequences of IRS-1 gene exon 1 were amplified by PCR.The amplified product was sequenced and identified.The polymorphism of IRS-1 gene was analyzed and compared with that of NCBI. Results Two SNPs were detected from the amplified fragment of Zhongguonongda miniature pig strains.The SNP of C2870G changed the amino acid from Ala to Gly on the locus of 956. Conclusion The two SNPs found in Zhongguonongda miniature pig strains but not in Bama and Wuzhishan miniature pig strains may be due to their genetic diversity.
