  • 摘要: 目的: 研究人类白血病多药耐药细胞株HL60/ADM对8种常用化疗药的耐药性及环孢菌素A (CsA)对其耐药的逆转作用。方法: MTT比色法检测细胞耐药性及CsA对细胞耐药性的影响,流式细胞仪检测细胞内柔红霉素(DNR)的浓度。结果: HL60/ADM对DNR、ADM、VCR、Har、VP16、MIT交叉耐药,对DNR耐药性最强,对Acla和Ara-c基本不耐药。CsA使HL60/ADM细胞内药物浓度增加,而对HL60的胞内药物浓度基本无影响。结论: HL60/ADM对Acla基本不产生耐药性,阿克拉霉素可能成为治疗白血病和克服白血病多药耐药性的重要药物。CsA对HL60/ADM的耐药性有部分逆转作用。

     

    Abstract: Objective: To investigate the chemoresistance of human leukemia multidrug resistant cell line HL60/ADM and the reversal effect of CsA. Methods: MTT assay was used to evaluate chemoresistance and CsA s reversal effects. Accumulation of DNR was evaluated by flow cytometry (FCM). Results: HL60/ADM is cross resistant to DNR、ADM、VCR、Har、VP 16 and MIT, but not to Acla and Ara-c. CsA increases the intracellular DNR accumulation of HL60/ADM, but not to sensitive cell line HL60. Conclusion: HL60/ADM cell line is resistance to eight chemodrugs. But to Acla, the resistance is very small. This indicates that Acla may become one of the most important drugs in treating leukemia and overcoming the multidrug resistance. CsA can reverse the chemoresistance of HL60/ADM, so it is the important reversal agent of multidrug resistance.
