肝硬变患者TGF-β1基因在外周血中表达及与PⅢP血清水平的关系

Transformation growth factor β1 gene expression in the peripheral blood of patients with cirrhosis and its relation to PⅢP

  • 摘要: 目的:为验证肝纤维化动物模型和慢性肝病患者肝组织中,转化生长因子(TTGF-β1)的mRNA表达增强。作者分析肝硬变患者外周血TGF-β1mRNA和血清中有活性的TTGF-β1 蛋白及其结果与型前胶原N端肽(PⅢP)血清水平间的关系。方法:肝硬变患者50例,健康自愿者8人,取其外周血,分离单核细胞(PBMC),提取总RNA,用巢式RT-PCR扩增TTGF-β1 mRNA并测定血清中TTGF-β1 和PⅢP的水平。结果:分析表明患者外周血单核细胞(PBMC)中TTGF-β1 mRNA有高阳性率的表达。血清中TTGF-β1 与PⅢP的水平均明显高于对照。结论:研究结果证实肝硬变患者TTGF-β1 基因在外周血中表达增强,与PⅢP血清水平相关,表明TTGF-β1 可能在肝纤维化、胶原合成中起重要作用

     

    Abstract: Objective: The aims of the present study were to determine the expression of transformation growth factor-β1 (TGF-β1) mRNA and TGF-β1protein in the peripheral blood and its relation to serum levels of procollagen type Ⅲ N terminal peptide (PⅢP) in the patients with cirrhosis. Methods: Peripheral blood samples were obtained from 50 patients with cirrhosis and 8 normal healthy volunteers. Total RNA was extracted from PBMC and TGF-β1mRNA was amplified from total RNA by nested RT PCR. Serum levels of TGF-β1and PⅢP were determined. Results: TGF-β1mRNA could be detected in the peripheral blood mononuclear (PBMC) with high positive rate and serum levels of TGF-β1and PⅢP were significantly higher than controls.Conclusion: These results confirm the increased expression of TGF-β1and the positive correlation between TGF-β1and PⅢP in the patients with cirrhosis. And our results support the suggestion that TGF-β1may be a significant factor in the increased collagen systhesis of human fibrosis.
