兔急性肺损伤中性粒细胞弹性蛋白酶变化及氯喹的保护作用

Changes of neutrophil elastase and the protective effects of Chloroquine in exdotoxin-induces acute lung injury in rabbits

  • 摘要: 目的:用内毒素(ET)静注复制兔急性肺损伤(ALI)模型,探讨ALI的发病机制,并寻找有效的干预药物。方法:日本大耳白兔24只随机分为A组(对照组)、B组(损伤组)和C组(损伤+干预组)。A组静注生理盐水对照,B组按600μg/kg剂量静注ET复制ALI模型,C组静注同等剂量ET后按5 mg/kg剂量静注氯喹进行干预。分别测定各组静注药物前、后0.5 h、1 h、2 h、4 h动脉血气分析、丙二醛(MDA)、超氧化物歧化酶(SOD)和NE的变化,比较3组之间的差异。结果:注射ET后B组氧合指数、MDA和NE值明显升高,SOD降低,C组也有类似变化,但程度较B组轻。3组比较差异显著。结论:NE活性增强是ALI发生的重要机制,应用氯喹后血清中NE活性明显降低,氯喹对内毒素所致ALI有一定的保护作用。

     

    Abstract: Objective: To study the changes of neutrophil elastase(NE)of blood serum,blood gas analysis in endotoxin(ET)-induces acute lung injury(ALI) in rabbits by intravenous injection,and to observe the protective effects of Chloroquine in order to reveal the mechanism of ALI and to find out effective therapeutic medicine for ALI in the clinic.Methods: 24 Rabbits were randomly assigned to three groups: group A(control group),group B(ET group),and group C(ET and chloroquine group).ALI was induced by intravascular injection of ET(600 μg/kg).The rabbits of group C were injected by chloroquine after ET injection.The rabbits of group A were injected by normal sodium.The contents of NE,malondialdehyde(MDA),superoxide dismutase(SOD) in blood and blood gas analysis were measured before injection and after injection 0.5 h,1 h,2 h,4 h in every group.The results were analyzed by SPSS 13.0.Results: NE and MDA are higher in group B than that in group A.PaO2/FIO2,SOD in group B are lower than that in group A.The results of group C are similar with group B.But the extent of change in group C is lower than that in group B.The differences with each group are significant.Conclusion: The action of NE enhancement is one of the mechanisms of ALI,Chloroquine can depress the action of NE and it has protective effect to endotoxin-induced ALI.
