新生小鼠耳蜗体外培养的扫描电镜观察

SEM observation of post-natal mice cochleas cultivated in vitro

  • 摘要: 目的:了解新生小鼠耳蜗在体外条件下存活状况,为体外急性实验提供组织模型。方法:对出生1~3d小鼠耳蜗以体外悬浮培养方法,并用扫描电镜对其外毛细胞的结构和形态进行观察。结果:采用这种方法培养的外毛细胞,在存活的20h内,其细胞形态和结构无明显改变。结论:耳蜗悬浮培养对短期离体外毛细胞的研究不失为稳定可靠的方法,而扫描电镜也补充证实为一种有效、简便的观察手段

     

    Abstract: Objective: To observe cochleas of post-natal mice cultivated in vitro and establish a tissue model for acute experiment.Methods: The free-floating method was used to cultivate the cochleas of 1 to 3 days post-natal mice in vitro. The structure and morphology of outer hair cells were investigated with scanning electron microscope (SEM). Results: The structure and morphology of outer hair cells remained unchanged during 20 hours.Conclusion: The free floating method was optimal for the short-term research of outer hair cells in vitro and the SEM was proved to be an effective, convenient investigating method.
