一种快速纯化单克隆抗体的方法——阴阳双重离子交换层析法

A rapid method for preparing purified McAb——The anion and cation exchange chromatography

  • 摘要: 本文介绍了一种纯化单克隆抗体的方法——阴阳双重离子交换层析法。该方法利用单克隆抗体具有带电均一性的特点,将阴离交换树脂和阳离子交换树脂集于一个层析柱内.使层析柱既可吸附阳离子也可以吸附阴离子。这样处于兼性离子状态的待纯化单抗在层析柱中走的最快,而其它一些等电点不同于待纯化单抗的杂蛋白分子则带有正电荷或负电荷,可被柱中的离子交换树脂吸附,在层析柱走的较慢,从而与待纯化单抗分离。我们用这种新型的快速柱层析法纯化了抗结肠癌3B3单克隆抗体。其单抗纯度达到90%以上,取得了较理想的效果。

     

    Abstract: This report introduced a new simple method for preparing purified monoclonal antibodies (McAb),which could be called as the anion and cation exchange chromatography.For the molecules of monoclonal antibodies were uniform in charge,so the chromatography column that contains anion resin and cation resin was produced.In this condition,the McAb which is in facultative ion state can flow fast in column,and the other proteins whose isoelectric point is not equal to the McAb’s take positive charge or negative charge and can be absorbed by the resin among the chromatography column,so these proteins flow slower in the column.As the result, the McAb can be separated from ascites fluid.We purified 3B3 McAb against human colon carcinoma by this method.The purity of 3B3 McAb is more than 90%.
