奥普利侬对细菌性腹膜炎致大鼠膈肌疲劳的影响

Effects of Olprinone on rat diaphragmatic fatigability induced by CLP

  • 摘要: 目的: 观察预防性腹腔注射奥普利侬对大白鼠细菌性腹膜炎致膈肌疲劳肌张力的影响。方法: 用肓肠结扎术加穿孔(CLP)方法制成大白鼠腹膜炎模型。分组为(n=10):Ⅰ.假手术对照组;Ⅱ.CLP组;Ⅲ.CLP+10mg·kg-1奥普利侬(Olprinone)组;Ⅳ.CLP+20mg·kg-1奥普利侬组。实验时,首先用10、20、50、100Hz (刺激电压为6V,时间为1s)直接刺激肌肉并记录数值;然后,采用6V、2ms单刺激直接刺激肌肉,并记录峰颤张力(PTT)、达峰颤搐时间(TPT)和半量松弛时间(1/2RT)值。通过公式将记录的肌张力转换成单位面积所产生的肌张力值。结果: 与对照组相比,各种刺激所引起的Ⅱ、Ⅲ、Ⅳ组膈肌单位面积产生的肌张力显著降低(P<0.05)。Ⅲ、Ⅳ组与Ⅱ组相比,两者之间没有显著差异(P>0.05)。而6V、2ms单刺激所引起的Ⅱ、Ⅲ、Ⅳ组TPT和1/2RT改变与对照组相比没有明显变化(P>0.05),PTT值明显下降(P<0.05)。结论: 细菌性腹膜炎可使膈肌发生疲劳,预防性单次给予奥普利侬不能缓解大白鼠CLP腹膜炎引起的膈肌疲劳。

     

    Abstract: Objective: to evaluate the effect of Olprinone on the contractility of fatigued diaphragms in rats induced by cecal ligation and puncture(CLP). Methods: Rats were divided into four groups of 10 each. In group Ⅰ, the rats were only performed with artificial operation. In group Ⅱ, Ⅲ, Ⅳ, diaphragmatic fatigue was induced by CLP. Olprinone were injected into abdomen cavity with the dose of 10mg/kg or 20mg/kg in group Ⅲ and Ⅳ. During the experiment, the diaphragmatic muscles were first stimulated by the electric at 10Hz, 20Hz, 50Hz, 100Hz(6V, continues 1s) and the contractility was detected; then the diaphragmatic muscles were stimulated by single stimulation with 6V current continuing 2ms and the Peak Twitch Tensicn(PTT), Time to Peak Tension(TPT), Half relaxation time(1/2RT) were detected. Results: compared with artificial operation group, the contractility of the other three groups were decreased obviously (P<0.05); but there were no difference between the fatigued diaphragms induced by CLP with or without pre-treating with Olprinone(P>0.05). Also there were no difference in TPT and 1/2RT induced by 6V, 2ms single electric stimulation among all these four groups (P>0.05). conclusion:fatigued diaphragms could be induced by bacterial peritonitis, but pre-treating with single abdomen cavity injection Olprinone cannot relieve the rat diaphragmatic fatigability induced by CLP.
