丙泊酚复合咪唑安定对再灌注损伤PC12细胞的保护作用

Protection of propofol combined with midazolam on cultured PC12 cells impaired by ischemia and reperfusion

  • 摘要: 目的: 观察静脉麻醉药丙泊酚与苯二氮卓类药咪唑安定合用对缺血缺氧及再灌注PC12细胞损伤的保护作用,并探讨其可能的机制。方法: 分为缺血缺氧及再灌注损伤组(IP);丙泊酚12.4μmol.L-1组(P);咪唑安定3.3μmol.L-1(M1)、10μmol.L-1(M2)、30μmol.L-1组(M3);丙泊酚12.4μmol.L-1与咪唑安定3.3μmol.L-1(PM1)、10μmol.L-1(PM2)、30μmol.L-1(PM3)合用组。通过测定存活细胞的吸光度值(A570nm,MTT比色法)判断细胞损伤程度。并观察细胞内钙离子浓度的变化。结果: ①与IP组比较,P、M1、M2、M3组A570nm值明显升高(P<0.01),且M组呈剂量依赖性。②PM1、PM2、PM3组成活细胞数较M1、M2、M3组进一步升高,且PM2、PM3组成活细胞数与P组相比也显著升高(P<0.01)。③与IP组相比,P、M1、M2组Ca2+i均显著下降(P<0.01),PM1、PM2处理组Ca2+i进一步下降,并与P、M1、M2相比呈显著性差异(P<0.01)。结论: 丙泊酚和咪唑安定对缺血缺氧及再灌注 PC12细胞损伤均有保护作用,合用时保护作用进一步加强;这一作用至少部分与抑制细胞内钙超载有关。

     

    Abstract: Objective: To observe the effects of the combined administration of propofol and midazolam on PC12 cells impaired by the mimic ischemia and reperfusion(IR),and to investigate the possible mechanism.Methods: The PC12 cells impaired by IR were grouped as following;the ischemia and reperfusion(IP) group,propofol 12.4μmol·L-1 group,midazolam(3.3μmol·L-1,10μmol·L-1,30μmol·L-1)groups,propofol 12.4μmol·L-1 combined with midazolam(3.3μmol·L-1,10μmol·L-1,(30 μmol·L-1)) groups.The degree of impairment was measured by detecting the value of A570nm.Also the i was detected of the group IP and those treated by propofol(12.4μmol·L-1),midazolam(3.3μmol·L-1,10μmol·L-1),or both of them.Results: ①Compared with the IP group,the values of A570nm increased significantly in all groups treated by propofol and midazolam(P<0.01).Moreover,the values of A570nmincreased related to the concentration of midazolam.②The co-administration of propofol and midazolam could further increase the values of A570nm than which of midazolam and propofol(P<0.01).③Compared with IP group,i was decreased in the other treatment groups(P<0.01).Andi of the co-administration groups further decreased than which groups treated with propofol or midazolam alone(P<0.01).Conclusion: Both of propofol and midazolam can protect the PC12 cells against the impairment of IP,and the effects can be strengthen when they are administrated at the same time.The mechanism is not clear but at least partially relate to the inhibition of the overload ofi.anesthetics,intravenous;tranquilizing agents;hypoxia-ischemia,brain
