松花粉对D-半乳糖致亚急性衰老小鼠模型肾脏mtDNA的保护作用

Protective effect of pine pollen on renal mitochondrial DNA in a model of mice with subacute aging induced by D-galactose

  • 摘要: 目的 探讨松花粉对衰老模型小鼠肾脏线粒体DNA(mtDNA)的保护作用及其延缓衰老的机理。方法 将30只雄性昆明小白鼠随机分为青年对照组、衰老模型组、松花粉治疗组;衰老模型组、松花粉治疗组采用每天5%D-半乳糖溶液100mg/kg颈背部皮下注射,复制亚急性衰老小鼠模型;青年对照组给予0.9%氯化钠注射液;松花粉治疗组在造模的同时每天给予750mg/kg的松花粉灌胃;青年对照组与衰老模型组以等体积的0.9%氯化钠注射液灌胃。连续50d后观察各组肾脏病理改变,并检测肾脏组织mtDNA的相对含量、肾脏组织SOD活性及其MDA的含量。结果 松花粉能够改善衰老小鼠肾脏的病理改变,并能降低衰老小鼠肾脏组织mtDNA的相对含量,提高肾脏组织SOD活性及降低其MDA含量(P<0.05)。结论 松花粉可能通过减轻肾脏的氧化损伤,从而保护肾脏mtDNA延缓肾脏的衰老。

     

    Abstract: Objective To study the protective effect of pine pollen on renal mitochondrial DNA in a model of mice with subacute aging and its mechanism. Methods Thirty male Kunming mice were randomly divided into young control group,aging model group,and pine pollen group(n=10).Mice in the aging model and pine pollen groups received subcutaneous injection with 5% D-galactose(100mg/kg) daily to induce a subacute aging model of mice,mice in the young control group were given normal saline,and mice in the pine pollen group were given pine pollen(750mg/kg) daily.Enema was performed with normal saline in mice of the young control and aging model groups on the day when the model was established for 50 days.Then,pathological changes in kidney were then observed,and relative level of mtDNA and MDA and activity of SOD in kidney tissues were measured. Results Pine pollen ameliorated the pathological changes,reduced the relative mtDNA and MDA level in kidney,and increased the SOD activity in the aging mice(P<0.05). Conclusion Pine pollen can delay the kidney aging by reducing its oxidative damage and protecting its mtDNA.
