阿司匹林和奥美拉唑对大鼠胃黏膜损伤及血液NO、TNF的影响

Effect of aspirin and omeprazole on gastric mucosa injury and nitric oxide and tumor necrosis factor level in blood of rats

  • 摘要: 目的 研究阿司匹林及奥美拉唑对不同月龄大鼠胃黏膜和血液NO、TNF含量影响及作用机制。方法 月龄3个月和22个月Wistar大鼠各18只,分别随机分为损伤组、保护组和对照组。损伤组用100mg/kg阿司匹林灌胃,保护组用20mg/kg奥美拉唑灌胃,对照组生理盐水灌胃。检测血NO、TNF含量、NOS、iNOS及cNOS活力。Guth和whittle法分别比较各组大鼠胃黏膜损伤情况。结果 老龄损伤组损伤指数和whittle评分高于非老龄损伤组大鼠的损伤指数和whittle评分(P<0.05)。损伤组比对照组和保护组大鼠的TNF、NO含量及iNOS活力升高,cNOS活力下降(P<0.05)。胃黏膜损伤指数与iNOS活力、NO含量及TNF含量呈正相关,与cNOS活力呈负相关。结论 老龄大鼠的胃黏膜对损伤的敏感性增加,修复能力下降;阿司匹林可降低cNOS活力,升高iNOS活力,升高NO和TNF含量而损伤胃组织;奥美拉唑可降低炎性介质TNF含量和改变NO含量发挥保护作用。

     

    Abstract: Objective To study the effect of aspirin and omeprazole on gastric mucosa injury and nitric oxide (NO) and tumor necrosis factor (TNF) level in blood of rats at different ages and its mechanism. Methods Eighteen healthy Wistar rats at the age of 3 and 22 months were randomly divided into normal control group, aspirin injury group and omeprazole protection group, 6 rats in each group. Rats in aspirin injury group, omeprazole protection group and normal control group underwent gastric lavage with aspirin, omeprazole and normal saline, respectively. Levels of NO and nitric oxide synthase (NOS) and activity of NOS, iNOS and cNOS were measured. Scores for gastric mucosa injury in different groups of rats were compared with Guth and Whittle methods. Results The Guth and whittle scores for injuries in group of aged rats were higher than those group of non-aged rats (P<0.05). The TNF and NO level and the iNOS activity were higher in aspirin injury group than in normal control and omeprazole protection groups. However, the activity of cNOS was lower in aspirin injury and omeprazole protection groups than in normal control group. The gastric mucosa injury was positively correlated with the activity of iNOS and the level of NO and TNF, but negatively correlated with the activity of cNOS. Conclusion Gastric mucosa of aged rats is more sensitive to injury factors while its repair ability decreases. The mechanism by which aspirin injures gastric mucosa of rats may be related with increasing iNOS activities, NO and TNF levels, and decreasing cNOS activities. Omeprazole protects gastric mucosa against injury by decreasing the TNF and NO level.
