磨损颗粒数量对人工关节假体-骨界面巨噬细胞激活影响的实验研究

Experimental study on the relationship between macrophage activation andthe amount of wear particles

  • 摘要: 目的:探讨磨损颗粒数量对人工关节假体骨界面巨噬细胞(MФ)激活的影响。方法:以正常人髋关节滑膜细胞系为模型,借助激光扫描共聚焦显微镜(CLSM),动态监测分别加入1.0 mg/ml和1.5 mg/ml(W/V)Ti合金和CoCr合金颗粒悬液后巨噬细胞样细胞(MCs)内游离Ca2+浓度(Ca2+i)的变化。结果:滴加不同浓度的两种颗粒悬液后1 h内,MCs内Ca2+i可出现1~6次脉冲样升高,其脉冲次数、间隔时间、波动幅度等均与颗粒种类及浓度无关(P>0.05),唯Ti合金颗粒低浓度组出现Ca2+i脉冲的潜伏期较高浓度组长(P<0.05)。空白对照组及成纤维细胞样细胞(FCs)不出现类似反应。结论:假体骨界面MФ激活本身与磨损颗粒数量无关,但活化MФ的数量可能与颗粒数量有关,这是关系到界面是否发生骨溶解及其程度的重要中间环节。

     

    Abstract: Objective: To study the effect of the amount of wear particles on macrophage activation at the bone implant interface.Methods: 1.0mg/ml and 1.5mg/ml (W/V) Ti alloy and CoCr alloy particles suspension was added respectively into synoviocyte system of normal hip joint in vitro. The iof macrophage like cells(MCs) was monitored dynamically by confocal laser scanning microscope (CLSM). Results: Calcium channels of MCs membrane opened for 1-6 times and caused rapid and transient pulses of iin cytoplasm within 1 hour. There were no significant differences between the times, interval and range of the pulse under the stimulation of different particles and different amount of particles (P>0.05) But the incubation period of 1.0mg/ml Ti alloy particles group was significantly longer than that of 1.5mg/ml Ti alloy particles group (P<0.05). MCs in control group that was stimulated by Hanks’did not appear such reaction Conclusions: Activation of macrophages at boneimplant interface do not correlate with the amount of wear particles while the number of activated macrophages may be relevant to the amount of particles.
