二羰基/L-木酮糖还原酶单克隆抗体的鉴定与应用

Characterization of monoclonal antibody against DCXR and its application

摘要: 目的: 抗人二羰基/L-木酮糖还原酶(dicarbonyl/L-xylulose reductase,DCXR)的单克隆抗体(mAb)的初步鉴定与应用。方法: 通过免疫荧光染色法对DCXR mAb进行亚细胞定位,通过免疫沉淀法对DCXR mAb进行初步应用研究。结果: DCXR mAb定位于肝脏的线粒体,通过免疫沉淀法该mAb可以从线粒体总蛋白中去除相应的蛋白DCXR。结论: 成功制备了抗人DCXR的mAb,为DCXR的研究提供了有力的工具。

Abstract: Objective: To characterize the monoclonal antibody,mAb,against dicarbonyl/L-xylulose reductase（DCXR）for its application. Methods: DCXRmAb was detected by immunofluorescence staining. MAb was used to isolate DCXR by immunoprecipitation. Results: DCXRmAb was localized in mitochondria of liver and could deplete DCXR from total lysate in mitochondria by immunoprecipitation. Conclusion: Specific mAb against DCXR can be used as a powerful tool in investigation of DCXR functions and distribution.
