浅低温体外循环手术前后基因表达研究中内参基因的选择

Selection of internal control genes in gene expression study before and after hypothermal cardiopulmonary bypass

  • 摘要: 目的 筛选浅低温体外循环(cardiopulmonary bypass,CPB)手术前后基因表达变化研究的内参基因。方法 通过阅读文献选择基因表达研究中常用的11个管家基因(housekeeping genes,HKGs),采用Beacon Designer软件设计PCR引物;收集4例浅低温CPB辅助手术患者CPB前、体温最低点、CPB末以及术后1d、3d、7d的静脉血24份,提取总RNA,采用SYBR Green I实时荧光定量RT-PCR法检测各样品中11个HKGs的循环阈值;对所得循环阈值进行转换,采用geNorm程序对候选基因的表达稳定性和所需内参基因数量进行分析。结果 11个HKGs稳定性排序为:UBC<RPLP0<PGK1<18S-rRNA<PPIA<GAPDH<ACTB<TBP<B2M<EEF1A1/HPRT1;该研究需要的最佳内参基因数量为3个,即EEF1A1、HPRT1和B2M。结论 确定了浅低温CPB手术前后11个HKGs中表达稳定性高的EEF1A1、HPRT1和B2M等3个基因为该条件下基因表达研究的内参基因。

     

    Abstract: Objective To select the internal control genes in gene expression study before and after hypothermal cardiopulmonary bypass(CPB). Methods Eleven human housekeeping genes(HKGs) were included in this study,and the primer pairs for their amplification were designed by Beacon Designer software. Venous blood samples were collected from 4 patients before CPB,at minimum body temperature,at end of CPB,and days 1,3 and 7 after CPB. After total RNA was extracted from blood samples,mRNAs were measured by SYBR Green I based real-time fluorescent quantitative RT-PCR and cycle threshold values were acquired. Finally,average expression stability value of the remaining HKGs and the optimal number of internal control genes for normalization were calculated and determined according to the cycle threshold values by geNorm VBA applet. Results The 11 HKGs according to their expression stability were UBC,RPLP0,PGK1,18S-rRNA,PPIA,GAPDH,ACTB,TBP,B2M and EEF1A1/HPRT1 in order. The last three genes were the optimal internal control genes for the research. Conclusion Three most stable genes,EEF1A1,HPRT1 and B2M,can be chosen as optimal internal control genes in gene expression study before and after hypothermal cardiopulmonary bypass.
