Abstract:
Objective:To study the effects of different cryoprotectants on human skin,in order to offer experiment gist for selecting the best cryoprotectants.
Methods:The skins to be cryopreserved were first treated with DMSO-propyleneglycol,trehalose-DMSO,respectively,to be compared with fresh skin.Then the histological structure of skin of different groups was observed and analyzed by pathological technology(immunohistochemical staining).Furthermore,we investigated the influence of trehalose on β1 integrin at gene level with RT-PCR.The viability of skin in five respective groups was evaluated using SDH assay.The experiments were carried out 7 days and 14 days after cryopreservation.
Results: The results of histological studies showed that β1 integrin could be well-protected by trehalose.The results of RT-PCR test and SDH assay also revealed that skin cryopreserved with trehalose-DMSO was as good as fresh skin,and better than other groups.
Conclusion:The results suggested that cryopreservation protocol-trehalose-DMSO is better than the traditional cryoprotectant for cryopreservation on β1 integrin of human skin.
