阿霉素磁性明胶微球制备及镇痛作用的研究

摘要: 目的: 研究阿霉素磁性明胶微球的制备,探讨阿霉素磁性明胶微球的镇痛作用。方法: 采用乳化-交联法制备阿霉素磁性明胶微球。28只雄性新西兰兔,随机分为假手术组、空白微球5 mg对照组、空白微球15 mg对照组、阿霉素明胶磁性微球5 mg组、阿霉素明胶磁性微球15 mg组,在外磁场作用下蛛网膜下腔注射微球,连续观察家兔下肢的电痛阈、运动功能的变化。结果: 制备的阿霉素磁性明胶微球最佳投料比为1∶15,磁吸附率为100%。微球外形圆整,分散性好。阿霉素120 m in释放90%以上。蛛网膜下腔注射15 mg阿霉素磁性微球后,家兔痛阈显著升高(P<0.01),持续30 d仍未见消失。所有家兔运动功能评分未见明显变化。结论: 制备的阿霉素磁性明胶微球缓释性好,磁响应性强,有“感觉-运动”分离作用,能有效镇痛。Abstract: Objective: To study preparation and characteristics of adriamycin(ADM) magnetic gelatin microspheres,and to investigate its analgesic effect.Methods: Adriamycin magnetic gelatin microspheres were prepared by emulsion-crosslinking.Twenty-eight rabbits were randomly divided into 5 groups sham operation group,gelatin microsphere controlled group(5 mg and 15 mg),and adriamycin microsphere group(5 mg and 15 mg).Under the effect of magnet,microspheres were infused into cavitas subarachnoidealis of rabbit.The pain threshold of electronic stimulates and motor function of rabbits’ hinder limbers were observed continuously.Results: The optimal ratio of adriamycin magnetic gelatin microsphere was 1∶15.The magneto-adsorption rate of the microspheres was 100%.The outline of microsphere was regular.The dispersivity and fluidity were perfect.The percent of drug release from adriamycin magnetic gelatin microspheres was over 90% during 2 hours.Adriamycin magnetic gelatin microsphere at dose of 15mg injected into subarachnoid space,caused obvious rise of pain threshold(P<0.01)for more than 30 days,but not in motor function.Conclusion: The prepared adriamycin magnetic gelatin microsphere has good properties of drug-targeting and drug sustain releasing.It can cause the dissociation of kino-sense,so has obviously analgesic effect.
