口服补镁对实验性老年2型糖尿病大鼠胰岛素受体亲和力的影响

Effect of oral magnesium supplementation on insulin receptor affinity in elderly rats with experimental type 2 diabetes mellitus

  • 摘要: 目的 观察口服补镁对老年2型糖尿病大鼠胰岛素受体亲和力的影响。 方法 采用给老年2型糖尿病大鼠模型口服补镁的方法,测定肝细胞胰岛素受体亲和力,并应用高胰岛素、正葡萄糖钳夹技术测定稳态下葡萄糖输注率、空腹血糖、空腹胰岛素及血浆镁离子浓度等指标。 结果 高剂量补镁组(2g/kg)高、低亲和力胰岛素受体结合常数分别为(4.83±0.09)×108/mol和(1.12±0.17)×106/mol,高、低亲和力胰岛素受体结合容量分别为(4.46±0.16)×1010/mg Pr、(8.32±0.27)×1013/mg Pr,与糖尿病对照组相比各组数据均显著升高(P<0.05);同时,高剂量补镁组空腹血糖、空腹胰岛素及胰岛素敏感指数HOMA-IR明显下降,稳态下葡萄糖输注率显著升高。 结论 镁补充可提高2型糖尿病大鼠胰岛素受体亲和力,降低胰岛素抵抗。

     

    Abstract: Objective To observe the effect of oral magnesium supplementation on insulin receptor affinity in elderly rats with type 2 diabetes mellitus(DM). Methods After oral magnesium was supplemented to type 2 DM model of elderly rats,liver cell insulin receptor affinity was assayed,serum levels of fasting glucose,insulin and magnesium,and stable glucose infusion rate were measured with the euglycaemic-hyperinsulinaemic clamp technique. Results The constant of high and low affinity insulin receptor banding was significantly higher while the capacity of high and low affinity insulin receptor banding was significantly lower in high dose magnesium(2g/kg) supplementation group than in DM control group(4.83±0.09)×108/mol Pr vs(1.12±0.17)×106/mol Pr,(4.46±0.16)×1010/mg Pr vs(8.32±0.27)×1013/mg Pr,P<0.05).The serum levels of fasting glucose and insulin,and insulin sensitivity index(HOMA-IR) were significantly lower while the stable glucose infusion rate was significantly higher in high dose magnesium(2g/kg) supplementation group than in DM control group. Conclusion Oral magnesium supplementation can improve the insulin receptor affinity and reduce the insulin resistance in elderly rats with type 2 DM.
