反复+Gz暴露对自发性高血压大鼠靶器官损害的组织学观察

Repeated +Gz exposure induced damage to target organs in rats with spontaneous hypertension

  • 摘要: 目的 观察反复正加速度(+Gz)暴露后自发性高血压大鼠(spontaneously hypertensive rats,SHR)心、脑、肾、主动脉的病理改变。 方法 14只8周龄SHR及14只8周龄Wistar Kyoto大鼠(wistar kyoto rats,WKY)随机分为4组,SHR+Gz暴露组,SHR对照组,正常WKY+Gz暴露组,正常WKY对照组,每组各7只。+Gz暴露采取模拟空战动作模式连续进行,依次为+5 Gz/10 s、+9 Gz/10 s、+5 Gz/10 s、+9 Gz/10 s、+5 Gz/10 s和+9 Gz/10 s,G增长率为1 G/s。休息20 min后,再按上述模式重复暴露1次,连续进行7 d暴露。对照组大鼠不进行离心机暴露。所有大鼠在实验结束后立即采集心、脑、肾、主动脉标本,进行HE染色光镜观察,并对左心室心肌质量指数(left ventricle mass index,LVMI)进行测量。 结果 SHR大鼠LVMI明显高于正常WKY大鼠(P<0.01),+Gz暴露SHR大鼠LVMI明显高于对照SHR组(P<0.01)。光镜观察两组WKY大鼠靶器官未见明显组织学损害表现;SHR+Gz暴露组与SHR对照组大鼠存在心肌、肾脏、大脑皮质损害表现,SHR+Gz暴露组损害程度重于SHR对照组。 结论 +Gz暴露可以加重SHR靶器官的组织学损害程度。

     

    Abstract: Objective To observe the repeated +Gz exposure induced damage to heart, brain, kidneys and aorta in rats with spontaneous hypertension (SH). Methods Fourteen SH rats and 14 Wistar Kyoto rats (WKY) at the age of 8 weeks were randomly divided into SH +Gz exposure group, SH control group, WKY +Gz exposure group and WKY control group (7 in each group). The rats were exposed to +5 Gz/10 s, +9 Gz/10 s, +5 Gz/10 s, +9 Gz/10 s, +5 Gz/10 s and +9 Gz/10 s respectively by simulating air combat maneuver (SACM), which were repeated after the rats had a 20 min rest, total for 7 days. Samples taken from the heart, brain, kidneys and aorta of the rats at the end of exposure were stained with HE and observed under light microscope. The left ventricular mass index (LVMI) was measured. Results The LVMI was significantly higher in SH rat groups than in WKY rat groups (P<0.01), and was significantly higher in SH +Gz exposure group than in SH control group. No significant target organ damage was observed in WKY +Gz exposure group and WKY control group under optical microscope. The damage to cardiac muscle, kidneys and cerebral cortex was severer in SH +Gz exposure group than in SH control group although it was observed both in the two groups. Conclusion +Gz exposure can aggravate the damage to target organs in SH rats.
