骨髓基质细胞移植对大鼠脊髓损伤后环氧化酶-2和血管内皮生长因子表达的影响

Effect of bone marrow stromal cells transplantation on expressions of cyclooxygenase-2 and vascular endothelial growth factor in rats following spinal cord injury

  • 摘要: 目的 观察骨髓基质细胞 (bone marrow stromal cells, BMSCs) 移植对大鼠脊髓损伤后环氧化酶-2 (cyclooxygenase-2, COX-2) 和血管内皮生长因子 (vascular endothelial growth factor, VEGF) 蛋白表达的影响。 方法 大鼠随机分为3组:A为假手术组, B为细胞培养基 (DMEM) 对照组, C为BMSCs移植组。B组和C组大鼠进行脊髓压迫损伤模型制作。脊髓损伤后, B组和C组大鼠分别给予DMEM培养液或BMSCs脊髓损伤周围区注射。术后应用Western Blot方法检测COX-2、VEGF蛋白在损伤脊髓组织中的表达变化。 结果 COX-2、VEGF蛋白在A组大鼠未损伤脊髓组织中均低水平表达。B组与A组相比, 脊髓损伤后COX-2蛋白表达水平明显增加 (P<0.01) , VEGF蛋白表达水平则短暂显著增加 (P<0.05) 。C组与B组相比, COX-2和VEGF蛋白表达水平均显著增加。 结论 BMSCs移植能够增加脊髓损伤后COX-2、VEGF蛋白的表达。

     

    Abstract: Objective To observe the effect of bone marrow stromal cells(BMSCs) transplantation on expression of cyclooxygenase-2(COX-2) and vascular endothelial growth factor(VEGF) protein in rats following spinal cord injury(SCI). Methods Adult rats were randomly divided into group A(sham operation group),group B(DMEM control group),and group C(BMSCs transplantation group).A rat spinal cord compression injury model was established.Rats in groups B and C were injected with BMSCs or DMEM after SCI.Expression of COX-2 and VEGF protein in rats following SCI was detected by Western blot. Results The expression level of COX-2 and VEGF protein was low in group A.The expression level of COX-2 and VEGF protein was significantly higher in group B than in group A and in group C than in group B(P<0.01,P<0.05). Conclusion BMSCs transplantation can up-regulate the expression of COX-2 and VEGF protein in rats following SCI.
