标本保存温度和保存时间对流式自然杀伤细胞和B细胞数量检测的影响

Effect of sample storage temperature and time on NK cells and B-lymphocytes detected by flow cytometry

  • 摘要:  目的   检测标本保存时间和保存温度, 为流式自然杀伤 (natural killer, NK) 细胞和B细胞检测标准化操作提供科学依据。  方法   采用四色流式细胞术分析健康体检人群外周血NK细胞和B细胞数量百分比, 检测保存时间与保存温度对实验结果的影响。  结果   未经处理的全血标本常温 (18℃) 保存后检测结果稳定, 低温、高温及长时间保存都引起检测结果降低;全血标本经荧光抗体染色后, 用多聚甲醛固定可在4℃条件下保存1周。  结论   流式细胞分析法检测NK细胞和B细胞, 若标本不能及时处理宜18℃保存并在96 h内检测;处理后的样本可在4℃条件下保存1周。

     

    Abstract:  Objective   To study the effect of storage temperature and time on the detection of NK cells and B-lymphocytes.  Methods   NK cells and B-lymphocytes in peripheral blood were detected by 6-color flow cytometry.The effect of storage temperature and time on the detection of NK cells and B-lymphocytes was observed.  Results   No change was found in the number of NK cells and B-lymphocytes in whole blood samples stored at 18℃.The number of NK cells and B-lymphocytes decreased after stored at low or high temperature.The whole blood samples that were fixed with paraformaldehyde and stained with fluorescent antibody could be stored for 7 days.  Conclusion   Flow cytometry can detect NK cells and B-lymphocytes.The samples can be stored at 18℃ for 96 h when it is not able to treat them in time.The treated samples can be stored at 4℃ for 7 days.
