抗表皮生长因子受体单抗联合X线照射对人鼻咽癌细胞株CNE-2的作用

Effect of combined anti-EGFR and X-ray irradiation on human nasopharyngeal carcinoma cell line CNE-2

  • 摘要: 目的 观察抗表皮生长因子受体(epidermal growth factor receptor,EGFR)尼妥珠单抗(h-R3)、西妥昔单抗(C225)分别联合X线照射对人鼻咽癌细胞株CNE-2的作用。 方法 采用流式细胞仪测定CNE-2细胞表面EGFR表达情况;采用PCRDNA测序技术筛选K-ras野生型CNE-2细胞;采用MTS法检测细胞增殖情况;采用克隆形成实验拟合细胞存活曲线;通过流式细胞仪分析细胞周期分布及细胞凋亡变化。 结果 在调节CNE-2细胞克隆存活及凋亡方面,C225联合X线照射组的作用优于h-R3联合X线照射组(P<0.05);在细胞增殖及细胞周期方面,h-R3联合X线照射组与C225联合X线照射组之间差异无明显统计学意义(P>0.05)。 结论 h-R3与C225均能提高X线照射对CNE-2细胞的抗肿瘤作用,C225对该细胞株杀伤作用略优于h-R3。

     

    Abstract: Objective To study the effect of anti-EGFR mAbs, nimotuzumab (h-R3) and cetuximab (C225), in combination with X-ray irradiation on human nasopharyngeal carcinoma cell line CNE-2. Methods Expression of surface EGFR in CNE-2 cells was detected by flow cytometry (FCM). Wild K-ras CNE-2 cells were detected by PCR-DNA sequencing. Proliferation of CNE-2 cells was assayed by MTS. Cell survival curves were plotted by clone formation experiment. Cell-cycle distribution and apoptosis were analyzed by FCM. Results The effect of combined C225 and X-ray irradiation was better than that of combined h-R3 and X-ray irradiation on the survival and apoptosis of CNE-2 cells (P< 0.05). However, no significant difference was observed in their effect on cell cycle distribution and proliferation (P> 0.05). Conclusion Both h-R3 and C225 can enhance the anti-tumor effect of X-ray irradiation on CNE-2 cells. The effect of C225 is better than that of h-R3 on CNE-2 cells.
