Methylation of PCDH10 gene in prostate cancer tissue and its clinical significance

  • 摘要: 目的 检测PCDH10基因在前列腺癌组织中的甲基化状态,并分析其临床意义。 方法 应用甲基化特异性PCR (methylation-specific PCR,MSP)技术检测PCDH10基因在3株前列腺癌细胞系、1株前列腺上皮细胞、13例前列腺增生组织和40例前列腺癌组织样本中的甲基化情况,同时分析40例前列腺癌患者的临床资料。 结果 3株前列腺癌细胞系中有2株检出PCDH10基因甲基化;40例前列腺癌样本中24例(60%)检出PCDH10基因甲基化,13例前列腺增生组织中未检出PCDH10基因甲基化。PCDH10基因出现甲基化患者与未出现甲基化患者相比,在年龄、前列腺特异性抗原(prostate specific antigen,PSA)和临床分期等指标的差异无统计学意义(P>0.05),但在Gleason评分的差异存在统计学意义(P<0.05)。 结论 PCDH10基因在前列腺癌组织中甲基化程度较高,高Gleason评分的前列腺癌可能具有更高的PCDH10基因甲基化率,提示PCDH10基因甲基化可能与前列腺癌的发生发展有关。

     

    Abstract: Objective To detect the methylation of PCDH10 gene in prostate cancer (PCa) tissue and analyze its clinical significance. Methods Methylation of PCDH10 gene was detected in 3 PCa cell lines, 1 prostate epithelial cell line, 13 hyperplasic prostatic tissue samples and 40 PCa tissue samples by methylation-specific PCR. Clinical data about the 40 PCa patients were analyzed. Results Methylation of the PCDH10 gene was detected in 2 out of the 3 PCa cell lines and in 24 out of the 40 PCa tissue samples with a detection rate of 67.7% and 60.0%, respectively. However, no methylation of the PCDH10 gene was detected in the 13 hyperplasic prostatic tissue samples. No significant difference was found in the age, prostate specific antigen (PSA), and clinical stage between those with or without methylation of the PCDH10 gene (P< 0.05). The Gleason score was significantly higher in patients with methylation of the PCDH10 gene than in those without methylation of the PCDH10 gene (P< 0.05). Conclusion The incidence of PCDH10 gene methylation is rather high in PCa patients. The methylation rate of PCDH10 gene is related with the higher Gleason score, suggesting that PCDH10 gene methylation is related with the pathogenesis and progression of PCa.
