维生素C对小鼠丙泊酚麻醉后血清及脑组织抗氧化能力的影响

Effects of vitamin C on antioxidant activity of serum and brain tissue in mice after propofol anesthesia

  • 摘要: 目的 探讨维生素C对小鼠丙泊酚麻醉后血清及脑组织抗氧化能力的变化。 方法 取50只6 ~ 8周龄、体质量20 ~24 g昆明雄性小鼠,随机分为0.9%氯化钠注射液对照组、丙泊酚(propofol,PPF。以下简称P)80 mg/kg组、P 60 mg/kg +Vc 67 mg/kg(简称P60+Vc67,下同)组、P55+Vc100组、P50+Vc200组(n=10),采用腹腔注射给药的方式,给予小鼠丙泊酚与维生素C不同剂量配比的混合药物,待小鼠翻正反射消失1 min后,进行眼眶后静脉丛采血及提取脑组织,利用分光光度计及酶标仪测定各组血清及脑组织中丙二醛、超氧化物歧化酶的含量及总抗氧化能力。 结果 P60+Vc67组、P55+Vc100组、P50+Vc200组中小鼠血清中的丙二醛值均低于P80组;P60+Vc67组、P55+Vc100组、P50+Vc200组中小鼠血清中的超氧化物歧化酶、总抗氧化能力值均高于P80组。P55+Vc100组及P50+Vc200组小鼠脑组织中的丙二醛值均低于P80组;P60+Vc67组、55+Vc100组及P50+Vc200组小鼠脑组织中的超氧化物歧化酶、总抗氧化能力值均高于P80组。 结论 维生素C可使昆明鼠丙泊酚麻醉后血清及脑组织总抗氧化能力提高。

     

    Abstract: Objective To explore the antioxidant capacity of propofol combined vitamin C by observing the change of level of MDA, SOD, T-AOC in serum and brain tissue of KM mice. Methods Fifty male KM mice with age of 6-8 weeks and weight of 20-24 kg were randomly divided into fve groups: 0.9% sodium chloride injection control group, 80 mg/kg propofol (PPF) group, 67 mg/kg P60+Vc group, 100 mg/kg P55+Vc group and 200 mg/kg P50+Vc group, 10 in each group. Rats were injected intraperitoneally with different dose of mixture of propofol and vitamin C, and after 1 min of loss of righting refex (LORR), blood sample from orbital venous and brain tissues were collected, and MDA, SOD, T-AOC in serum and brain of all groups were measured by ELISA. Results Compared with PPF80 group, the MDA level of serum in P60+Vc67, P55+Vc100, P50+Vc200 group was lower (P< 0.05), while the SOD, T-AOC level of serum in P60+Vc67, P55+Vc100, P50+Vc200 group was higher (P< 0.05). And the MDA level of serum in P55+Vc100, P50+Vc200 group was lower (P< 0.05), the SOD, T-AOC level of serum of P60+Vc67, P55+Vc100, P50+Vc200 group was higher (P< 0.05). Conclusion Vitamin C can enhance the total antioxidant capacity after KM mice undertaking propofol, which indicates that vitamin C can increase the antioxidant capacity of KM mice after propofol anesthesia.
