大鼠皮肤不同烧伤深度对早期白蛋白渗出量的影响

Effect ofdifferent burndepths in skin of rats on early albumin exudation

  • 摘要: 目的 探讨大鼠皮肤不同烧伤深度对早期白蛋白渗出量的影响。 方法 在16只成年雄性SD大鼠腹部两侧分别用恒温焊台的金属头在78℃下造成皮肤中度和深度烧伤,烧伤后5 min内从尾静脉注射0.5%偶氮蓝溶液(1 ml/250 g),3 h后分别在不同烧伤深度和未烧伤部位取材放入甲酰胺中,在50℃条件下振荡24 h析出偶氮蓝,测量甲酰胺的OD值,通过大鼠皮肤中偶氮蓝渗出量表示蛋白的渗出量。 结果 不同偶氮蓝浓度的甲酰胺溶液与其OD值呈现高度的相关性;未烧伤、中度烧伤、深度烧伤皮肤组织偶氮蓝含量分别为(29.4±6.4) μg/g,(146.4±28.7) μg/g,(308.6±25.2) μg/g,大鼠中度烧伤和深度烧伤皮肤白蛋白渗出量分别增加了约5倍和10倍。 结论 烧伤后早期大量白蛋白通过烧伤创面丢失到体外,且白蛋白的丢失随着烧伤深度的加重明显增加。

     

    Abstract: Objective To investigate the influence ofdifferent burndepth in skin of rats on early albumin exudation. Methods With the help of metal head of constant temperature welding machine, either side of the abdomen was heated in 16 adult male SD rats at 78℃ respectively which caused skin moderate anddeep burns. 0.5% Evans blue (1 ml/250 g) was injected from tail vein at 5 minutes after burned. Samples were taken indifferentdepth of burn and non-burned areas 3 h later and each sample was placed in formamide and incubated for 24 h in a shaking water bath at 50℃ to extract Evans blue from the tissue and the OD values of formamide samples were measured. The Evans blue extravasation in rat's skin represented the amount of albumin exudation. Results Formamide solution ofdifferent concentrations of Evans blue and its OD value showed a highdegree of correlation. The content of Evans blue of nonburned, moderate burn anddeep burn skin tissues were (29.4±6.4) μg/g, (146.4±28.7) μg/g, (308.6±25.2) μg/g, respectively. The amount of albumin exudation in the rat's skin of moderate burn anddeep burn increased about 5 and 10 times, respectively. Conclusion In the early stage of burn injury, a large amount of albumin loses in vitro, and the loss of albumin increases with the increase of thedepth of burn.
