低聚乙二醇富马酸酯/氧化石墨烯复合水凝胶对大鼠骨髓间充质干细胞成骨分化的调控作用

Effects of oligo poly (ethylene glycol) fumarate/graphene oxide on osteogenic differentiation of bone marrow mesenchymal stem cells in rats

  • 摘要: 目的 探讨低聚乙二醇富马酸酯oligo poly (ethylene glycol) fumarate,OPF/氧化石墨烯复合水凝胶(graphene oxide,GO)对大鼠骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)成骨分化的调控作用。 方法 通过物理共混方式将氧化石墨烯掺杂在低聚乙二醇富马酸酯水凝胶中,并将骨髓间充质干细胞包裹其中共培养,成骨诱导3 d、7 d、14 d 和21 d后,通过组织学染色(HE染色、Von Kossa染色以及茜素红染色)和免疫荧光染色检测复合材料对BMSCs形态及其对成骨分化的影响。 结果 组织学染色及免疫荧光染色发现,BMSCs在水凝胶内良好生长并增殖,随着成骨诱导时间的延长,矿化结节以及成骨基因表达逐渐升高,且相比OPF水凝胶,OPF/GO水凝胶组促进BMSCs向成骨细胞分化的作用更强。 结论 OPF/GO水凝胶具有良好的生物相容性,且利于BMSCs向成骨细胞分化。

     

    Abstract: Objective To investigate the effects of oligo poly (ethylene glycol) fumarate/graphene oxide (OPF/GO) on differentiation of bone marrow mesenchymal stem cells (BMSCs) in rats. Methods OPF/GO hydrogel were prepared via physical mixing.Then BMSCs were encapsulated in OPF/GO hydrogel and co-cultured in osteogenic medium.After 3, 7, 14 and 21 days, the morphology and osteogenic differentiation of BMSCs were detected by histological staining (HE staining, Von Kossa staining and alizarin red staining) and immunofluorescence staining. Results Cells encapsulated in composite hydrogel were stained for histological examination and immunofluorescence staining at different time points (3, 7, 14 and 21 days), and it was found that BMSCs grew and proliferated well in the hydrogels.With the extension of osteogenic induction, mineralized nodules and osteogenic gene expression gradually increased.Moreover, compared with OPF hydrogel, the OPF/GO could improve the osteogenenic differentiation of BMSCs significantly. Conclusion OPF/GO hydrogel has good biocompatibility and it can induce the osteogenenic differentiation of BMSCs effectively.
