山莨菪碱联合新斯的明对大鼠胆肠吻合术后炎症反应的影响

Effect of anisodamine combined with neostigmine on inflammatory response in rats after bilioenteric anastomosis

  • 摘要: 目的 探讨山莨菪碱联合新斯的明对大鼠胆肠吻合术后炎症反应的影响。 方法 选用SD大鼠30只,随机分为5组,每组6只。包括正常组、空白对照组(0.9%氯化钠注射液1 ml)、低剂量组(山莨菪碱12.5 mg/kg联合新斯的明25μg/kg)、正常剂量组(山莨菪碱25 mg/kg联合新斯的明50μg/kg)和高剂量组(山莨菪碱50 mg/kg联合新斯的明100μg/kg)。确定分组后正常组立即处死,各用药组大鼠于术后第1天即开始给予相应药物腹腔注射,连续7 d后处死。比较各组大鼠体质量及炎症指标(CRP、TNF-α、IL-1β)。 结果 各用药组大鼠均顺利完成二期胆肠吻合术,无术中死亡。高剂量组1只大鼠于第2天注射药物后死亡,其他各组无死亡。胆肠吻合术后1周,空白对照组、低剂量组、正常剂量组和高剂量组大鼠的TNF-α、IL-1β高于正常组大鼠(P< 0.05)。正常剂量组大鼠的CRP为(2.7±0.1) mg/L,TNF-α为(1.8±0.2) pg/ml,IL-1β为(69.5±6.3) pg/ml,均明显优于空白对照组大鼠(P< 0.05)。低剂量组大鼠各炎症指标均与空白对照组大鼠无统计学差异。 结论 山莨菪碱联合新斯的明能有效降低大鼠胆肠吻合术后炎症反应,最佳药物浓度为山莨菪碱25 mg/kg联合新斯的明50μg/kg,低剂量药物作用效果不明显,而高剂量药物有引起动物死亡的风险。

     

    Abstract: Objective To study the effect of anisodamine combined with neostigmine on inflammatory response in rat after bilioenteric anastomosis. Methods A total of 30 SD rats were randomly divided into 5 groups, 6 in each group, including normal group, control group, low dose group, normal dose group and high dose group. Rats in normal group were sacrificed right now after grouping. Rats in normal dose group received anisodamine (25 mg/kg) combined with neostigmine (50μg/kg) on the first day after operation by intraperitoneal injection, and continued for 7 days. Low dose group rats received half of the normal dose (anisodamine for 12.5 mg/kg and neostigmine for 25μg/kg), high dose group rats received 2 times of the normal dose (anisodamine for 50 mg/ kg and neostigmine for 100μg/kg), and rats in control group were given normal saline. The weight and inflammation indexes (CRP, TNF-α, IL-1β) of rats in different groups were compared at 1 week after operation. Results The operation was successfully completed in each rat. No rats died except one in the high dose group after the second day of injection. One week after operation, TNF-α, IL-1β of rats in control group, low dose group, normal dose group and high dose group were significantly higher than normal group (P< 0.05). While, there were no significant differences in weight and inflammation indexes of rats in low dose group and control group. The results of CRP, TNF-α and IL-1β of normal dose group were (2.7±0.1) mg/L, (1.8±0.2) pg/ml and (69.5±6.3) pg/ml, which were significantly lower than those of control group (P< 0.05). Conclusion Anisodamine combined with neostigmine can effectively reduce the systemic inflammatory response in rats after bilioenteric anastomosis. The optimal drug concentration is anisodamine (25 mg/kg) combined with neostigmine (50μg/kg), the drug effect of low dose is not significant, and high dose of the drugs may have a risk of causing the death of animals.
