miR-199-3p在兔膝骨关节炎软骨下骨中的表达及其意义

Expression of miR-199-3p in subchondral bone from rabbits with knee osteoarthritis

  • 摘要: 目的 探讨MicroRNA-199-3p (miR-199-3p)在兔膝骨关节炎软骨下骨中的表达。 方法 24只新西兰大白兔随机分为前交叉韧带切除(anterior cruciate ligament transaction,ACLT)组及对照组,每组12只。ACLT组右膝前交叉韧带切除制造骨关节炎模型,对照组不作何处理,分别于术后4周、8周、12周取材。切片行甲苯胺蓝染色,样本进行软骨下骨分离后,运用实时定量PCR检测不同时间点软骨下骨中miR-199-3p的表达情况。 结果 ACLT组术后4周即可见软骨退变,8周和12周时退变进一步加重,甲苯胺蓝染色显示基质丢失、软骨变薄、软骨下骨裸露。各组软骨下骨中均能检测到miR-199-3p的表达。ACLT组中软骨下骨中miR-199-3p的表达量明显低于对照组,而MMP-3与IL-1β的表达量明显升高,且各时间点之间有统计学差异(P< 0.05)。 结论 miR-199-3p的低表达水平与骨性关节炎的严重程度密切相关。

     

    Abstract: Objective To detect the expression of microRNA-199-3p (miR-199-3p) in subchondral bone of rabbit model with knee osteoarthritis (OA). Methods Twenty-four New Zealand white rabbits were randomly divided into two groups:anterior cruciate ligament transaction (ACLT) group (n=12) and sham group (n=12). OA was surgically induced in the right knee of rabbits in ACLT group. The rabbit knees were evaluated at 4, 8 and 12 weeks after ACLT by gross observation and histological examination. After getting the specimens, we segregated subchondral bone, and the expression of miR-199-3p was tested by qRT-PCR. Results Rabbits in ACLT group showed typical OA pathological changes after 4 weeks, such as loss of surface integrity, fissuring and mircocracks, loss of cartilage and these symptoms aggravated at 8 and 12 weeks after operation. The expression of miR-199-3p could be detected in all subchondral bone with a decreasing trend over time, and it was significantly lower in ACLT group comparing with sham group, while the expression of MMP-3 and IL-1β were higher in ACLT group than sham group with significant difference at each time point (P< 0. 05). Conclusion Our findings indicate that low expression of miR-199-3p is closely related with the development and progression of OA.
