肾移植术后供受者尿蛋白质组差异蛋白分析

Urinary proteome analysis in recipients and donors after kidney transplantation

  • 摘要: 目的 比较肾功能稳定的肾移植供受者的尿蛋白质组变化,并分析差异蛋白生物学功能。 方法 选择5对于2010 -2014年在解放军总医院行肾移植术的肾功能稳定的肾移植受者及其相应供者的尿液样本,采用LC-MS/MS蛋白质组学技术进行蛋白质定量,并对筛选的差异蛋白进行生物信息学分析。 结果 5对供受者间共存在12个共同的差异蛋白,其中8个蛋白在肾移植受者尿液中表达上调:二磷酸腺苷核糖基转移酶3、碳酸酐酶4、视黄醇结合蛋白4、基底细胞黏附分子、过氧化物酶6、乳酸脱氢酶B、丝氨酸蛋白酶抑制剂A1和丝氨酸蛋白酶抑制剂A3。肾移植受者尿液表达中下调蛋白4个:细胞液泡蛋白质分选因子4B、囊泡运输体1、Bro1包涵域蛋白及三肽氨基肽酶1。 结论 移植肾功能正常的受者与健康供者的尿蛋白质组有差异,这些差异蛋白大多定位于细胞外环境,主要与物质转运、能量代谢及免疫反应等多种生物学过程有关。

     

    Abstract: Objective To explore the difference in urinary proteome between recipients and their corresponding donors after kidney transplantation, then analyze the bioinformation of these distinct proteins. Methods Urine samples from 5 recipients and their corresponding donors who underwent renal transplant in Chinese PLA General Hospital from 2010 to 2014 were analyzed by LC-MS/MS technology. Then the bioinformation of the distinct proteins were analyzed. Results There were 12 distinct proteins demonstrated remarkble variance in recipients and donors. Among them, 8 proteins showed up-regulation in recipients'urine, including Ecto-ADP-ribosyltransferase 3, Carbonic anhydrase 4, Retinol-binding protein 4, Basal cell adhesion molecule, Peroxiredoxin-6, L-lactate dehydrogenase B chain, Alpha-1-antichymotrypsin andAlpha-1-antitrypsin. While another 4 proteins showed down-regulation in recipients' urine, including Vacuolar protein sorting-associated protein 4B, Vacuolar protein sortingassociated protein VTA1 homolog, BRO1 domain-containing protein BROX and Tripeptidyl-peptidase 1. Conclusion The urinary proteome of recipients is different from donors after kidney transplantation. The distinct proteins are located in extracellular space and mainly related to biological processes like immune response or energy metabolism.
