猕猴子宫腺肌病动物模型建立初探

Establishment of animal models of adenomyosis in rhesus monkeys

  • 摘要:
      目的  探讨自体内膜种植法建立猕猴子宫腺肌病动物模型的可行性和建模成功率。
      方法  选择健康雌性猕猴6只,随机分为手术建模组(5只)和空白对照组(1只)。手术建模组猕猴行开腹手术,取出子宫内膜种植到子宫前壁肌层内,于术后第9个月行超声引导下穿刺活检,进行组织病理学检查判断建模效果。
      结果  手术建模组猕猴中4只术后一般情况良好、无严重并发症,其中1只猕猴于术后6个半月时死亡(尸检证实死亡原因为吞食猴毛导致上消化道梗阻)。死亡猕猴大体标本及3只手术组猕猴穿刺活检组织经病理学检查镜下子宫肌层内见子宫内膜腺体;1只手术组猕猴和对照组猕猴子宫前壁穿刺活检组织镜下子宫肌层内未见子宫内膜腺体结构。建模成功率为80%(4/5)。
      结论  自体内膜种植法建立猕猴子宫腺肌病模型是一种可行、有效、成功率较高的建模方法,不同猕猴的个体差异是建模成功与否的重要因素。

     

    Abstract:
      Objective  To evaluate the feasibility and success rate of autologous endometrial transplantation method in establishing adenomyosis animal model in rhesus monkeys.
      Methods  Six healthy female rhesus monkeys were randomly divided into operation modeling group (n=5) and blank control group (n=1). For the experimental group, the endometrium was taken out through laparotomy and implanted into the anterior wall of the uterus. Ultrasound guided puncture biopsy was done for the pathological examination at 9 months after operation.
      Results  In the operation modeling group, four monkeys were in good condition without serious complications, and one monkey died at 6.5 months after operation (autopsy confirmed that the cause of death was upper gastrointestinal obstruction caused by swallowing monkey hairs). Endometrial glands were found in the myometrium of the dead monkey and the biopsy tissues of three monkeys in the operation group; endometrial glands were not found in the myometrium of one monkey in the operation group and the monkey in the control group; the modeling success rate was 80% (4/5).
      Conclusion  It is feasible to establish the uterine adenomyosis model of rhesus monkeys by autologous endometrial implantation, with high success rate. The individual differences of different monkeys are the important factors for the success of modeling.
