Expression of sirtuin 1 in epithelial cells of chronic rhinosinusitis without nasal polyps and its effects on TGF-β1-induced epithelial mesenchymal transition

      背景  慢性鼻窦炎是耳鼻喉科常见的慢性炎症性疾病。通常不伴鼻息肉的慢性鼻窦炎(chronic rhinosinusitis without nasal polyps,CRSsNP)更常见,其病因及发病机制尚不清楚。
      目的  研究组蛋白去乙酰化酶1(sirtuin 1,SIRT1)在CRSsNP上皮细胞中的表达,并探讨其对转化生长因子-β1(transforming growth factor-β1,TGF-β1)诱导鼻黏膜上皮-间质转化(epithelial-mesenchymal transition,EMT)的影响。
      方法  采用免疫组织化学染色、Real-time PCR、Western blot观察SIRT1在CRSsNP和正常对照筛窦黏膜上皮中的表达情况。对正常鼻黏膜上皮细胞进行原代培养,并予TGF-β1处理,通过Western blot检测SIRT1过表达对EMT相关蛋白(E-cadherin、α-SMA、N-cadherin、Vimentin)表达的影响。
      结果  与对照组相比,CRSsNP组SIRT1表达减少 (P<0.001)。TGF-β1下调鼻黏膜上皮中SIRT1的表达,同时E-cadherin表达减少,α-SMA、N-cadherin、Vimentin表达增加,这一效应可被SIRT1过表达所逆转。
      结论  SIRT1抑制TGF-β1诱导的鼻黏膜上皮EMT,可能在CRSsNP的病理生理过程中起重要作用。

     

    Abstract:
      Background   Chronic rhinosinusitis is a common chronic inflammatory disease. While the majority cases of this disease are not complicated with nasal polyps (chronic rhinosinusitis without nasal polyps, CRSsNP). However, the etiology and pathological mechanisms of CRSsNP remain unclear.
      Objective  To investigate the expression of sirtuin 1 (SIRT1) in epithelial cells of CRSsNP and determine its effects on TGF-β1-induced epithelial mesenchymal transition (EMT).
      Methods  Nasal epithelial cells (NECs) were collected from patients with CRSsNP and controls. SIRT1 expression was detected by means of Real-time PCR, immunohistochemistry and Western blot. In addition, human NECs were cultured under TGF-β1. The effects of SIRT1 on TGF-β1-induced EMT were investigated.
      Results  SIRT1was specifically down-regulated in patients with CRSsNP (P<0.001). TGF-β1 decreased SIRT1 expression in vitro. Furthermore, the expression levels of E-cadherin were down-regulated, while the expression levels of α-SMA, vimentin and N-cadherin was up-regulated, and SIRT1-overexpression could reverse this process.
      Conclusion  SIRT1 suppress TGF-β1-induced EMT in human NECs, indicating that SIRT1 may play important roles in the pathogenesis of CRSsNP.
