NDLIP对大鼠脑缺血再灌注损伤后血脑屏障的保护作用

Effect of noninvasive delayed limbischemic preconditioning on endothelial barrier antigen expression and blood-brain barrier permeability after ischemia/reperfusion injury in rats

  • 摘要: 目的 观察无创性肢体缺血预适应(noninvasive delayed limbischemic preconditioning,NDLIP)对大鼠脑缺血再灌注损伤后血脑屏障(blood brain barrier,BBB)的保护作用。 方法 30只雄性SD大鼠随机分成NDLIP组、缺血再灌注组和假手术组。NDLIP组、缺血再灌注组采用阻断大鼠一侧大脑中动脉(MCA)血流2h,再灌注24h制成局灶性脑缺血模型。于再灌注前,NDLIP组左后肢无创性3次缺血5min/再灌5min,1次/d,连续3d;缺血再灌注组和假手术组不给予NDLIP。再灌注24h后断头取脑,制作冰冻切片,免疫组化染色检测BBB内皮屏障抗原(endothelial barrier antigen EBA)、纤维蛋白原(fibrinogen)的表达,并测量各组梗死体积。 结果 缺血再灌注组(I/R)EBA表达5.3±1.24,假手术组EBA表达15.2±1.02,两组比较有统计学差异(P<0.01)。NDLIP组EBA表达11.61±0.98,较缺血再灌注组显著增加(P<0.01);缺血再灌注组fibrinogen表达8.60±3.12,假手术组fibrinogen表达1.30±0.46,两组比较有统计学差异(P<0.01)。NDLIP组fibrinogen表达5.56±2.58,较缺血再灌注组显著减少(P<0.01);NDLIP组梗死体积(218.05±112.14)mm3,缺血再灌注组梗死体积(250.68±129.76)mm3,两组比较有统计学差异(P<0.01)。 结论 NDLIP能显著增加脑缺血再灌注损伤后EBA表达,降低BBB通透性,缩小梗死体积,对BBB及脑缺血再灌注损伤具有保护作用。

     

    Abstract: Objective To observe the protective effect of noninvasive delayed limbischemic preconditioning(NDLIP) on blood brain barrier(BBB) after ischemia/reperfusion(I/R) injury in rats. Methods Thirty male SD rats were randomly divided into NDLIP group,I/R group,and sham operation group,10 in each group.A focal cerebral I/R injury model of rats was established by occluding the middle cerebral artery(MCA) for 2h and reperfusing for 24h.Rats in NDLIP group were preconditioned before ischemia by occluding the lateral femoral arteries 3 times at an interval of 5min and reperfusing for 5min,once a day for 3 days.The prefrontal cortex was removed from rats 24h after reperfusion and cut into sections after freezing.Rats in I/R and sham operation groups were not given NDLIP.Expressions of endothelial barrier antigen(EBA) and fibrinogen were detected with immunohistology staining and infarct volume was measured. Results The expression of EBA was 11.61±0.98,5.3±1.24 and 15.2±1.02,respectively,in NDLIP,I/R and sham operation groups(P<0.01).The expression of EBA was 11.61±0.98,8.60±3.12 and 1.30±0.46,respectively,in NDLIP,I/R and sham operation groups(P<0.01).The expression of fibrinogen was 5.56±2.58,8.60±3.12 and 1.30±0.46,respectively,in NDLIP,I/R and sham operation groups(P<0.01).The infraction volume was(218.05±112.14)mm3 and(250.68±129.76)mm3,respectively,in NDLIP and I/R groups(P<0.01). Conclusion NDLIP can significantly increase the EBA expression,decrease the BBB permeability and cerebral infarction volume after I/R injury,thus protecting BBB against I/R injury.
