热灼联合液氮冷冻建立兔中晚期股骨头缺血性坏死动物模型

Rabbit intermediate-late avascular necrosis of femoral head model induced by thermal injury in combination with liquid nitrogen freezing

  • 摘要: 目的 探讨热灼联合液氮冷冻法建立兔中晚期股骨头缺血性坏死(ANFH)动物模型的方法,为治疗研究提供适宜的实验对象。 方法 对20只5月龄日本大耳白兔采用电烙铁热灼联合液氮冷冻左侧股骨头负重面,不造成股骨头脱位,右侧对照,分别在术后1、2、4、6、8周进行股骨头标本的大体解剖学观察,X线、MRI检查和组织切片光镜观察。 结果 术后1周股骨头出现坏死,2周后出现修复迹象,4周时关节面出现塌陷,6周时塌陷加重股骨头明显变扁,8周时股骨头碎裂、缩小并出现骨性关节炎改变。 结论 热灼联合液氮冷冻法可用于建立兔中晚期ANFH动物模型,动物无死亡,造模成功率高。

     

    Abstract: Objective To study the rabbit intermediate-late avascular necrosis femoral head(ANFH) model induced by thermal injury in combination with liquid nitrogen freezing. Methods Twenty 5-month old Japanese big-ear rabbits were included in this study.A intermediate-late ANFH model was induced by thermal injury in combination with liquid nitrogen freezing without dislocation of hip joint to observe their gross anatomy 1,2,4,6 and 8 weeks after operation.Tissue sections were observed under optical microscopy and examined by X-ray and MRI. Results Necrosis of femoral head occurred 1 week after operation and was repaired 2 weeks after operation.Joint surface of femoral head was collapsed 4 weeks after operation,which was aggravated 6 weeks after operation and became flat and smaller with osteoarthritis at week 8 after operation. Conclusion Thermal injury in combination with liquid nitrogen freezing can be used to establish rabbit intermediate-late ANFH model with no death of rabbits and a high success rate.
