比较卡铂对乳腺癌细胞及血小板生成素作用下巨核细胞的影响

Effect of carboplatin on breast cancer cells and thrombopoietin on megalokaryocytes

  • 摘要: 目的 研究化疗药物卡铂对乳腺癌细胞和使用血小板生成素(thrombopoietin,TPO)的巨核细胞(MO7e)的影响。 方法 MTT法测定不同浓度的TPO对巨核细胞株MO7e作用24h、48h、72h的影响,作出增殖曲线。测定不同浓度卡铂对乳腺癌MDA-MB-453细胞和使用TPO的MO7e细胞作用24h、48h的抑制率,求出半数抑制浓度(IC50),比较两者的IC50。 结果 MO7e细胞增殖率随TPO浓度增大而增大,TPO浓度为256ng/ml时进入平台期。卡铂对MO7e细胞的IC50约为对乳腺癌细胞IC50的10倍。 结论 TPO可促进巨核细胞增殖,卡铂对乳腺癌细胞的抑制作用明显高于对使用TPO的巨核细胞的抑制作用。

     

    Abstract: Objective To study the effect of carboplatin on breast cancer cells and thrombopoietin(TPO) on proliferation of megalokaryocytes. Methods Effect of different TPO concentrations on megakaryocyte cell line,MO7e cells was detected by MTT assay 24,48,and 72h after MO7e cells were exposed to TPO.A proliferation curve of MO7e cells was plotted.Twenty-four and forty-eight hour inhibition rate of different carboplatin concentrations on breast cancer MDA-MB-453 cells and TPO on MO7e cells were calculated to measure and compare their half inhibition concentration(IC50). Results TPO increased the proliferation of MO7e cells in a conceetration-dependent manner and reached its plateau when at 256ng/ml.The IC50 of MO7e cells was 10-fold higher than that of breast cancer cells. Conclusion TPO can promote the proliferation of megakaryocytes.The inhibitory effect of carboplatin on breast cancer cells is significantly higher than that of TPO on megakaryocytes.
