Bcl2-siRNA increases sensitivity of human lung adenocarcinoma A549/DDP cells to cisplatin

  • 摘要: 目的 探讨Bcl2-siRNA对人肺腺癌顺铂耐药细胞株A549/DDP对顺铂敏感性的影响。 方法 在人肺腺癌顺铂耐药细胞株A549/DDP细胞中转染bcl2-siRNA,48h后加入不同浓度(0、3、6、9、12、15μg/ml)新鲜配置的顺铂溶液,药物作用48h后,用CCK-8法检测细胞生长抑制率。荧光实时定量PCR(Real time RT-PCR)检测A549/DDP细胞转染bcl2-siRNA后Bcl-2mRNA的变化,Western Blot检测A549/DDP细胞转染bcl2-siRNA后Bcl-2蛋白的变化。 结果 在A549/DDP细胞中转染bcl2-siRNA后,细胞Bcl-2 mRNA降低92.6%(P<0.05),Bcl-2蛋白表达也明显降低,同时细胞对顺铂的敏感性增加57.8%(P<0.05)。 结论 Bcl2-siRNA能降低A549/DDP中Bcl-2 mRNA及蛋白的表达,增加A549/DDP对顺铂的敏感性。

     

    Abstract: Objective To study the effect of Bcl2-siRNA on sensitivity of cisplatin-resistant human lung adenocarcinoma A549/DDP cells to cisplatin. Methods Forty-eight hours after Bcl2-siRNA was transfected with cisplatin-resistant human lung adenocarcinoma A549/DDP cells,a fresh cisplatin solution was prepared by adding different concentrations of cisplatin(0μg/ml,3μg/ml,6μg/ml,9μg/ml,12μg/ml,15μg/ml) into A549/DDP cells to measure their growth-inhibition rate by CCK-8 assay.Expression of Bcl-2 mRNA and protein in A549/DDP cells-transfected Bcl2-siRNA was detected by Real time RT-PCR and Western blot,respectively. Results The expression of Bcl-2 mRNA and protein in A549/DDP cells-transfected Bcl2-siRNA decreased 92.6%(P<0.05) while the sensitivity of A549/DDP cells to cisplatin increased 57.8%(P<0.05). Conclusion Bcl2-siRNA can decrease the expression of Bcl-2 mRNA and protein in A549/DDP cells,and increase the sensitivity of A549/DDP cells to cisplatin.
