蟾蜍灵对食管癌ECA109细胞凋亡的影响及机制探讨

Effect and mechanism of bufalin on apoptosis in human esophageal cancer ECA109 cells

  • 摘要: 目的 观察蟾蜍灵对食管癌ECA109细胞凋亡的影响并探讨其机制。 方法 MTT法检测蟾蜍灵对细胞的增殖抑制作用;流式细胞术分析细胞凋亡情况;Western-Blot法检测凋亡抑制蛋白survivin,caspase-3蛋白的表达。 结果 随着蟾蜍灵浓度及作用时间的增加,细胞存活率逐渐降低(P<0.05),细胞凋亡率明显增高(P<0.05);随着药物浓度增加,survivin蛋白表达逐渐下降,同时活化caspase-3。 结论 蟾蜍灵可诱导食管癌ECA109细胞凋亡,其机制可能与抑制survivin蛋白和活化caspase-3有关。

     

    Abstract: Objective To study the effect of bufalin on apoptosis of human esophageal cancer ECA109 cells and its mechanism. Methods Inhibitory effect of bufalin on proliferation of human esophageal cancer ECA109 cells was assayed by MTT.Apoptosis of human esophageal cancer ECA109 cells and expression of survivin and caspase-3 were detected by flow cytometry and Western blot,respectively. Results The survival rate of human esophageal cancer ECA109 cells decreased while the apoptotic rate of human esophageal cancer ECA109 cells increased with the increasing concentration and time of bufalin used(P<0.05).The expression level of survivin and the activation caspase-3 protein in human esophageal cancer ECA109 cells increased with the increasing concentration of bufalin(P<0.05). Conclusion Bufalin induces apoptosis of human esophageal cancer ECA109 cells possibly by down-regulating the expression of survivin and the activation of caspase-3.
