缺氧诱导因子-1α在胶质瘤中的表达及意义

Expression and clinical significance of hypoxia inducible factor-1α in gliomas

  • 摘要: 目的:研究缺氧诱导因子-1α(hypoxia inducible factor-l alpha,HIF-1α)在人脑不同级别胶质瘤中的表达,分析HIF-1α、血管内皮生长因子(vascular endothelial growth factor,VEGF)和微血管密度(microvascular density,MVD)的相互关系,探讨HIF-1α与胶质瘤病理分级、血管生成的关系。方法:采用免疫组化检测54例胶质瘤组织中HIF-1α及VEGF的表达情况,并以VIII-R-Ag作为标记计数微血管密度(MVD)。结果:54例胶质瘤组织HIF-1α的表达阳性率为61.1%(33/54),HIF-1α的表达强度和胶质瘤的病理分级呈正相关(χ2=14.24,P<0.05)。HIF-1α的表达和VEGF的表达呈正相关(χ2=4.02,P<0.05),HIF-1α和MVD呈正相关(t=3.22,P<0.05)。结论:HIF-1α与人脑胶质瘤组织中血管生成及病理分级密切相关,可作为一项有意义的预后指标。

     

    Abstract: Objective:To research the expression of hypoxia inducible factor-1α(HIF-lα) and its relationship with the pathological grades in gliomas.To evaluate the relationships of HIF-lα expression with VEGF and microvessel density(MVD) among the three and to investigate their association with pathological grades、angiogenesis of human gliomas.Methods: The expression of HIF-1α、VEGF and as well as the proteins associated with microvessel density in the gliomas were studied with SP immunohistochemical staining.Results: The positive expression of HIF-1α in human gliomas of 54 cases was 61.1%,which was related with the histological grade of tumor.The expression of HIF-1α was positively correlated with VEGF(χ2=4.02,P<0.05) and MVD(t=3.22,P<0.05).Conclusion: HIF-1α correlates positively to histological grade and angiogenesis.The expressions of HIF-1α can be used to assess the prognosis in glioma patients.
